ABSTRACT

Innovative and practical platform safety management is the foundation of the innovation and entrepreneurship for undergraduate students in universities and colleges, which plays a critical role in maintaining the stability of the university and society, and ensuring the safety of teachers and students. In this present paper, the complex situation and problems of innovative and practical platform safety management, such as the imperfect regulations, weak atmosphere, incomplete facilities, unprofessional management, etc., are discussed under the background of "innovation and entrepreneurship" in China. With the combination of the above problems and the actual situation of Chengdu Medical College, the constructions of the regulations, such as access regulation, certification regulation, check regulation, rewards and punishment regulation, responsibility regulation, and a series of standard operating procedures (SOP) have also been carried out. Furthermore, adequate safety education and proper safety drills have been performed during the whole process of safety management. In addition, a diverse and professional management team with putting more emphasis on the development of safety facilities has been formed. In conclusions, the practice and exploration on the safety management of undergraduate students' innovative and practical platform in this paper may provide a theoretical basis and practical value to optimize the safety management in local universities and colleges under the background of "innovation and entrepreneurship" in China.

ABSTRACT

Based on MOOC teaching mode and the combination of flipped classroom teaching theory, the role of internship practice is integrated into medical education and the combination of MOOC teaching and traditional teaching mode in medical humanities curriculum is explored in practice applications. This paper analyzed the cur-rent situation of medical humanities curriculum teaching operation under current situation; based on the teaching concept of MOOC,introduced the reform of teaching form and teaching method in blending teaching mode, and ex-plored the practical application and practical thinking of blending teaching mode. This mode not only plays a lead-ing role of teachers teaching, but also fully embodies the subjective status of students'learning, exercises the students'ability of thinking in practice at the same time, and is an effective mode to improve medical humanities curriculum teaching.

ABSTRACT

With the rapid development of medical genetics, online Medelian inheritance in man (OMIM) manifests a more and more important role in medical genetics teaching.Using the educational form combining ‘ classroom teaching, review writing and seminar’,‘ Query and use of OMIM ’was introduced into the education of medical genetics.Reality practice revealed that this educational practice maintained advanced and timely status of knowledge and deeply activated self-studying and independent thinking ability of students.

ABSTRACT

OBJECTIVETo explore the effects of CXCR7 knock-down by CXCR7-shRNA lentiviral vector on the proliferation and invasion of human hepatoma carcinoma cells in vitro.

METHODSCXCR7-shRNA lentiviral vector was transfected into hepatocellular carcinoma HCCLM3 cells. The changes in mRNA and protein expression of CXCR7 in the transfected cells were investigated using real-time PCR and Western blotting, respectively, and MTT assay was employed to assess the cell proliferation changes. In vitro adhesion assay and transwell chamber test were used to observe the adhesion and invasiveness of HCCLM3 cells, respectively.

RESULTSTransfection of HCCLM3 cells with CXCR7-shRNA lentiviral vector resulted in a significantly decreased expression of CXCR7 at both mRNA and protein levels (P<0.01) and obvious suppression of the cell proliferative activity (P<0.05). CXCR7-shRNA also significantly suppressed the invasiveness and adhesion of HCCLM3 cells (P<0.01).

CONCLUSIONCXCR7 knock-down can significantly inhibit the proliferation and invasiveness of human hepatoma carcinoma cells in vitro, suggesting the value of CXCR7 as a potential target for hepatoma carcinoma therapy.

ABSTRACT

Hypoxia-inducible transcription factor-1alpha (HIF-1alpha) is a "master" gene in the cell hypoxia response network. HIF-1alpha plays the key role on VEGF-dependent angiogenesis during tumor development. In this study, the siRNA expressive vector targeting HIF-1alpha was constructed. HepG2 cells were tranfected with an HIF-1alpha siRNA vector and exposed to a normoxic or hypoxic environment. mRNA and protein levels of HIF-1alpha and VEGF were detected by RT-PCR and Western blot at different cell culture environment. The mRNA and protein expression of HIF-1alpha was inhibited by RNA interference. The obvious decreases of mRNA and protein of VEGF was induced by HIF-1alpha gene knock-down. Our study indicated that HIF-1alpha regulated the expression of VEGF and hence promoted angiogenesis in liver cancer under hypoxic environment. The expression of VEGF was downregulated via HIF-1alpha gene knock-down. These results suggest that HIF-1alpha may be used as a good target gene for anti-angiogenesis in liver tumors.

ABSTRACT

Survivin is a member of inhibitor of apoptosis (IAP) family which has strong ability of antiapoptosis. Survivin is prominently expressed in transformed cell lines and in all the most common human cancers in vivo, but it is undetectable in terminally differentiated adult tissues. We designed a human survivin antisense oligonucleotides. RT-PCR and Western blot clearly showed that survivin mRNA and protein were decreased by this antisense oligonucleotide. The inhibition of SMMC-7721 cell proliferation was demonstrated by MTT assay. The inhibition rate (43.28% +/- 3.65%) was much higher than the inhibition rate (6.76% +/- 0.92%) of the control group. The apoptosis rate (33.68% +/- 2.89%) obtained by transfecting cells with the use of antisense oligonucleotide was higher than the apoptosis rate (14.12% +/- 1.24%) of the control group. And the result of TUNEL revealed that, when SMMC-7721 cells were exposed to homoharringtoine at low concentration and were transfected by antisense oligonucleotide, the apoptosis rate of the cells was significantly higher than that of other control group. These results indicate that antisense oligonucleotide may have the potential for selective tumor therapy in future.

ABSTRACT

Objective To investigate the association between the ATP-binding cassette transporter A1(ABCA1)gene polymorphisms and plasma high density lipoprotein cholesterol(HDL-C)level in Chinese general population. Methods M883I,R1587K and R219K polymorphisms of the ABCA1 gene were detected by polymerase chain reaction-restriction fragment length polymorphism technique in 376 unrelated Chinese healthy subjects. Results In the study population, the subjects with the KK1587 genotype had markedly lower HDL-C levels than those with RR1587 and RK1587 genotypes(P

ABSTRACT

Objective: To investigate the in vitro efficiency of two kinds of polylatic acid (PLA) immunomicrospheres: M1(hAFP158~166)and M2(hAFP218~226) in specific inducement of CTL and the cytotoxicity of CTL against hepatocellular carcinoma cells. Methods: Pripheral blood monocytes (PBMC) of HLA-A2+ healthy volunteers stimulated in vitro by peptide-loading microspheres were taken as effectors. The experiments were divided into three groups: control group, hAFP+ tumor cells group, and hAFP- tumor cells group. The specific cytolysis of target cells was assessed by 51Cr-release assay. Results: Both M1 and M2 induced proliferation of HLA-A2+ PBMC, forming visible clones. The effector cells induced by M1 and M2 both had an cytolysis rate over 75% for hAFP+ tumor cells, which was significantly higher than that for hAFP- tumor cells (P0.05). Conclusion: The two kinds of microspheres can both induce specific CTL in vitro, which can effectively kill hAFP+ tumor cells.
